个人简介 Personal Profile 猪繁殖与呼吸综合征病毒(PRRSV)引起机体免疫抑制和持续性感染,给养猪业造成损失巨大。我们前期实验发现重组 IFN-β能抑制 PRRSV 复制, 而PRRSV 通过非结构蛋白 nsp1α和 nsp11 拮抗其转录;所以抑制 I 型干扰素(IFN-β)产生可能是 PRRSV 抑制机体免疫应答的重要原因。最近又发现PRRSV通过调控4个宿主miRNA而抑制I 型干扰素转录,其中,PRRSV上调的miRNA-373通过靶向I型干扰素转录信号蛋白NFIA和NFIB(首次发现NFIA 和 NFIB 是 IFN-β 的转录因子,而且是拮抗PRRSV复制的宿主内源性蛋白)IRAK1、IRAK4、IRF1而逃逸宿主免疫;基于此,将进一步探讨PRRSV如何调控这些宿主miRNA而逃逸宿主的先天性免疫。   我们得到了nsp1α的单抗,其亲和力和滴度非常高,适合做PRRSV疫病的检测抗体,所以将开发基于nsp1α的PRRSV快速检测试纸。 研究方向Research Directions 病毒分子免疫学和病毒快速检测 2. 机电结构优化与控制 研究内容:在对机电结构进行分析和优化的基础上,运用控制理论进行结构参数的调整,使结构性能满足设计要求。1. 仿生结构材料拓扑优化设计, 仿生机械设计 研究内容:以仿生结构为研究对象,运用连续体结构拓扑优化设计理论和方法,对多相仿生结构(机构)材料进行2. 机电结构优化与控制 研究内容:在对机电结构进行分析和优化的基础上,运用控制理论进行结构参数的调整,使结构性能满足设计要求。1. 仿生结构材料拓扑优化设计, 仿生机械设计 研究内容:以仿生结构为研究对象,运用连续体结构拓扑优化设计理论和方法,对多相仿生结构(机构)材料进行整体布局设计。 整体布局设计。 科研项目 参加工作以来,主持国家自然科学基金1项、农业部重点实验开放课题1项、河南师范大学优秀青年培育项目1项和省教育厅项目1项,参加国家自然基金面上项目1项,省科技攻关1项,省教育厅项目1项。具体如下:1.        国家自然基金青年科学基金项目,31302073,猪繁殖与呼吸综合征病毒(PRRSV)非结构蛋白(nsp)1α和nsp11抑制β干扰素转录的分子机理研究,2014/01-2016/12,25万元,结题,主持。2.        农业部重点实验室开放课题,PKLAI20170605,miR-376b-3p促进猪繁殖与呼吸综合征病毒(PRRSV)复制的分子机理研究,2017.6-2019.6,10万元,在研,主持。3.        河南师范大学2016年度优秀青年科学基金项目,miR-376b-3p促进猪繁殖与呼吸综合征病毒(PRRSV)复制的分子机理研究,2017.6-2020.6,15万元,在研,主持。4.        河南省高等学校重点科研项目,16A180008,猪繁殖与呼吸综合征病毒N蛋白上调miR-373的分子机理研究,2016/01-2017/12, 5万元,在研,主持。5.        2018年度河南省自然科学基金,182300410077,抑制猪繁殖与呼吸综合征病毒(PRRSV)复制的宿主蛋白筛选及其机理研究,10万, 2018.1-2019.12 研究成果 以第1作者在Journal of Virology ,cellular ﹠ molecular immunology 等杂志发表SCI论文12篇,受理和授权国家发明专利5项;合作出版专著2本。具体如下:(1)         chen jing(#),shi xibao(#)(*)(共同一作及通讯作者),zhang xiaozhuan(#),zhang gaiping(*),MicroRNA 373Facilitates the Replication of Porcine Reproductive and Respiratory SyndromeVirus by Its Negative Regulation of Type I Interferon Induction,Journal of Virology,2017.01.01,91(3):e01311~e01316 (2)        Shi xibao(#)(*),zhang xiaozhuan(#),wang li,Zhang gaiping(*),Nonstructuralprotein 11 (nsp11) of porcine reproductive and respiratory syndrome virus(PRRSV) promoted PRRSV replication in MARC-145 cells,BMC Veterinary Research,2016. 12 (3)        shi xibao(#)(*),Xiaozhuan Zhang(#),Li Wang,Weiguo Li,Bo Jiang,Ruiguan Deng,Aiping Wang,Gaiping Zhang(*),Recombinant beta interferon could clear the low-doseinfected porcine reproductive and respiratory syndrome virus (PRRSV) but not tothe high-dose infected PRRSV in MARC-145 cells,ACTA VIROLOGICA,60(3), 290-297.
